Imaging in CNS Drug Discovery and Development Springer Verlag

Zdrowie, medycyna

Imaging is a key area of translational research that provides a unique bridge from the laboratory to the clinic as the same techniques and technologies can applied across the various phases of drug development. Neuroimaging is now central to research and drug development in the neurosciences since it can be used to detect the pharmacological and physiological consequences of drug action within the living brain. Today neuroimaging is used primarily as a biomarker to help optimize our decision-making processes. Neuroimaging tools and technologies can be used to select drug candidates, confirm they engage their targets, link occupancy to biology and then, in a translational fashion, be used to help design the best clinical studies that truly test therapeutic hypotheses. Neuroimaging today is all about the need to identify the best molecules, doses and hypotheses to advance into later stage long term extensive clinical trials that can often be confounded by high placebo response rates. Better decision making in early development allows resources to be focused on the drug candidates and therapeutic hypotheses that have the highest probability of success so that we can bring medical advances to patients as quickly as possible. Imaging in CNS Drug Discovery and Development was written by a group of preeminent international experts in the field of imaging.Topics covered include: * The role of imaging in drug development * Imaging approaches including structural, functional, molecular and chemical imaging * Imaging drug action including neuroreceptor mapping to examine drug occupancy characteristics, the involvement of specific neurotransmitter systems in CNS diseases and mechanisms of action * Functional mapping to provide pharmacodynamic evidence of central activity, "CNS fingerprinting" of the neuroanatomy of drug effects and disease drug interactions * Imaging disease states in preclinical and clinical domains

Drug Discovery in Pancreatic Cancer Springer Verlag

Medycyna > English Division

Pancreatic cancer is the fourth leading cause of cancer death in the United States. Every year, about 33,700 people in the United States will be diagnosed with pancreatic cancer and over 32,000 patients will die from the disease. The median survival of patients with advanced pancreatic cancer is about 6-months. This dismal picture of pancreatic cancer is mainly due to the lack of early diagnosis and effective treatment for patients with advanced disease. To increase the survival rate of pancreatic cancer patients, better tumor markers for diagnosis and new molecular targets for drug development are desperately needed. A lot of effort has been made in searching for pancreatic cancer-causing genes or genes associated with progression of malignant behavior in pancreatic cancer. As a result, alterations in the expression of several cancer-related genes have been identified in pancreatic tumors. The identification and characterization of these cancer-related genes have significantly increased our understanding of pancreatic cancer development, but unfortunately the treatment of pancreatic cancer has not advanced as much in the past 20 years. Over the past decade, tremendous advances have been made in the field of cancer drug discovery, particularly, in the area of molecular and genetic models and technologies. Many of those advanced models and technologies have been applied to the drug discovery processes for pancreatic cancer. In this book, a team of experts will describe the latest development in the application of these models and technologies in pancreatic cancer. The authors include basic researchers as well as clinicians who work in the front-line of the war against pancreatic cancer and have the first-hand experience on these cutting-edge tools and techniques. The book can be divided into two general areas: model systems and genomics and proteomics tools. In recent years there have been a lot of advances in the model systems for pancreatic cancer, including the further characterization of normal and cancerous pancreatic cell lines, the establishment of transgenic mouse models that recapitulate the initiation and progression of human pancreatic cancer, the development of a new xenograft model system for the evaluation of novel agents, and the establishment of a zebrafish pancreatic cancer model. The first four chapters of the book will be devoted to these models. The advances in genomics and proteomics research have made a major impact in cancer drug discovery. A number of these -omics-based tools and techniques have been applied in the pancreatic cancer drug discovery. Chapters 5-9 of the book will discuss techniques for genome-wide examination of gene expression, copy number, methylation, function and regulation. Chapters 10-11 will discuss in situ techniques for studying chromosomal and gene copy number abnormalities as well protein expression changes in cancer samples. Chapters 12-14 will focus on techniques for global examination of protein expression levels in biospecimens obtained from pancreatic cancer patients. Cancer drug discovery has become more and more target-centric.

Drug discovery and development is a challenging, expensive and time consuming field of research, requiring contributions from chemists, pharmacologists, toxicologists, clinicians, and practitioners. The ultimate goal is to generate a safe and biologically active drug which can stall, or even reverse, the pathological events that cause the disease condition. But in the search for the drug a host of tests and trials must be applied to evaluate the efficiency and safety of the newly developed molecule in the biological system. These trials or "screening methods" are critical. On their basis, the new molecule either becomes accepted for usage, or is discarded forever. Advances in drug research have forced the need for quicker, more automated screening methods, using molecular techniques applied in vitro, in vivo and in clinical systems. Researchers need to know the latest developments outside their own speciality. With this book, Professor Gupta has brought together in one coherent volume the most up to date developments of consolidated screening methods for biological systems. By paying attention to the practical techniques used in academia and the commercial pharmaceutical industry, "Drug Screening Methods" will enjoy a broad readership, serving both the professional community and the student of pharmacology.

Opis - Pierwsze na świecie świadectwo ofiary zbrodni honorowej. Miała siedemnaście lat i zakochała się: zhańbiła rodzinę. Więc rodzina wydała na nią wyrok śmierci... Pokochała go pierwszą miłością. Myślała, że się z nią ożeni. Ale ukochany zniknął, a ona odkryła, że jest w ciąży. A w jej świecie to najcięższa zbrodnia... Wyrok wydaje jej ojciec. Szwagier dokonuje egzekucji. Oblewa Souad benzyną i podpala...

Drug discovery and development is an expensive multi-disciplinary effort dependent on chemistry, structural biology, molecular and cell biology, pharmacology, physiology, statistics, computer sciences, engineering, medicine, law and business. It is a major challenge to incorporate all these fields into a single course, however it is also an opportunity to provide advanced students with a unifying context in which to apply knowledge from foundation courses. Rather than simply describing the drug discovery process, an essential goal of this text is to introduce the key decisions that are involved, the types of information needed to make these decisions, and the types of experiments used to obtain that information. In my teaching I have found that focusing on core concepts makes the subject accessible to a wider range of students and facilitates the integration of the knowledge into a context that is accessible and meaningful. This approach also serves to reinforce basic skills of research design and interpretation, and of data-driven decision-making skills relevant for a large number of potential learning and career directions. This book is intended to provide students with an understanding of the primary steps in the drug discovery and development process, key information needs associated with the decisions that govern the progression of projects from lab to clinic to market, and major research approaches used to provide information, support decision-making, and identify/prioritize candidate drug targets and candidate molecules. Along with a focus on the decision-making process, the text will emphasize the fundamental principles of science. Textboxes will highlight core concepts, techniques, and historic case examples, including examples from global infectious diseases such as malaria and sleeping sickness. End-of-chapter questions, a glossary and a math appendix will ensure that the text is accessible to upper-level undergraduates and graduate students.
